Subcellular and isoform-resolved spatial profiling with explicit annotation audits
Three items converge on spatial resolution but address distinct layers. Kintsugi estimates RNA composition in nucleus-poor compartments of subcellular spatial transcriptomics by borrowing information gene-by-gene from spatial neighbours and co-located genes. Spl-ISO-Seq2 with Spl-IsoQuant-2 and Spl-IsoFind extends spatial profiling to long-read isoforms at 500-nm resolution, reporting cell-type-specific spatial isoform variability across multiple brain cell types. AnnoAudit, a marker-based protocol, shows that a permissive labelling rule produced a spurious neuronal population in a widely used traumatic brain injury resource. The shared thread is that finer spatial and single-cell measurements require explicit validation of inferred composition, isoform and label assignments. What is new is the pairing of subcellular and isoform resolution with explicit auditing of annotation-derived artifacts.
