Computational methods sharpen spatial positioning and clone tracing in imaging assays

The common thread is computational and combinatorial improvement of spatial localization in imaging-based genomics. accumap applies statistical inference over predicted inter-bead distances to reconstruct bead positions in an optics-free, diffusion-based spatial transcriptomics workflow. SpaceBar takes a different route to resolution, pairing a 96-sequence synthetic barcode library with imaging-based spatial transcriptomics so that single-cell clone identity can be tracked alongside spatial gene expression. LEAR addresses localization error in multi-loci chromatin tracing as a post-processing step that requires no additional experiments, and is applied to enhancer biology. What is new across these items is that resolution is improved through inference, barcode design, or post-processing of existing measurements. Evidence is limited to one preprint and two published methods, so the shared thread reflects methodological direction rather than established consensus.
