Codon position and post-stop sequence context track translation efficiency and pausing
The dominant thread is that sequence features within coding regions, particularly codon position, are associated with translation behavior. The published POSCO study introduces a position-specific codon occurrence score linked to translational efficiency, tAI, ribosome occupancy and folding energy across the human transcriptome. A separate preprint trains a machine learning model to predict protein stability from N-terminal sequences of annotated and alternate isoforms, validates motifs experimentally, and applies the model to alternative isoforms. A yeast multi-omic preprint on glucose starvation examines cis-regulatory programs across multiple RNA regulatory layers, a theme less directly tied to codon-level mechanisms. What is new is the position-resolved codon metric applied at transcriptome scale, alongside sequence-based stability prediction for isoforms.
