Quantitative modeling links biomolecular structure to FRET readouts and dye properties
The common thread is the use of structural models to interpret fluorescence observables quantitatively. A review of quantitative modeling of fluorescence experiments on biomolecules covers models linking structure to FRET data, integrative modeling, dye representations, and dye property prediction. A bioRxiv preprint applies time-resolved FRET lifetime and anisotropy imaging, combined with brightness-based concentration estimation, to quantify MC4R oligomerization in an open-source framework. A published study uses HelixFold3 structural models and molecular dynamics to show that tryptophan burial accounts for variable fluorescence responses of IL-6 nanobody quenchbodies. What is new is the shift from qualitative fluorescence read-outs toward explicit structural and dye-level representations that make measurements interpretable at the molecular level.
